As the film begins, Arivudai Nambi (Vijay Antony) is killed by his rivals, who fear their position would be threatened if he were to make it big in politics. Decades later, his son Tamilarasan (Vijay Antony) is accidentally drawn into the murky world of behind-the-screens politicking when he is jailed due to admitting to have hit someone on the road. Two gangster-type baddies with political backing become his enemies, and this in turn brings him closer to Karunakaran (Thiagarajan), a political heavyweight who has his own ambitions. A minister sees Tamilarasan as an enemy because he sees in him Nambi, whom he had conspired to murder. Machinations and counter-machinations grow with the growing political ambitions of Tamilarasan, whose rivals won’t let him and his friend Anjana (Miya) live in peace. Tamilarasan boldly faces all this in his game of one-upmanship and realizes his dream of growing big in politics.

Aanusmita Sharma (Shahana Goswami), an employee of London-based company Barron Industries, introduces a new technology that allows objects from the digital world to enter the real world using the wireless transmissions from multiple devices. Shekhar Subramanium (Shah Rukh Khan), a game designer who also works at Barron Industries, has delivered a number of commercial failures; an irate Barron (Dalip Tahil) gives him his last chance to develop a successful game. To impress his sceptical son Prateek (Armaan Verma), and upon the request of his wife Sonia (Kareena Kapoor), Shekhar uses his son’s idea that the antagonist should be more powerful than the protagonist. His colleague, computer programmer Aanusmita uses Shekhar’s face as a model for that of the game’s protagonist G.One (Shah Rukh Khan) (Technically Good One and in Hindi Jeevan, which means life), while the shape-shifting antagonist Ra.One (Technically Random Access Version One and in Hindi Ravan, an ancient demon king) is made faceless. Another colleague, her boyfriend, Aakaashi (Tom Wu), implements the characters’ movements. The game, named Ra.One, contains three levels, the final level being the only one in which either character can be killed. Each character possesses a special device – the H.A.R.T (Hertz Amplifying Resonance Transmitter) which gives them their powers. Upon reaching the last level, the characters gain a gun with one bullet; the other character can be killed by this bullet but only if his H.A.R.T is attached.

Vandiyathevan sets out to cross the Chola land to deliver a message from the Crown Prince Aditha Karikalan. Kundavai attempts to establish political peace as vassals and petty chieftains plot against the throne. Unrest grips the land as a long-tailed comet arrives, signalling a time of turmoil.
Satya and Deepthi are lovers living in Visakhapatnam. A dreaded don, Veeru Bhai, lusts for Deepthi. When Deepthi’s parents are killed, she and Satya escape. They board a bus on route to Hyderabad, but the bus is stopped unexpectedly and boarded by three men: Veeru’s henchman Deva, Veeru’s brother Ajay, and a corrupt police inspector, Shravan. They kill Deepthi in front of Satya, and he suffers an almost-fatal stab wound. Before the three leave, they set fire to the bus, leaving Satya to his death after stabbing him. However, Dr. Shailaja saves Satya by giving him a new face and skin. Ten months later, Satya wakes up from his coma.
Ashok (Vishwa), Ani (Payal), Viji (Nanditha Jennifer), Divya (Laksha), Pravin (Kadhal Sukumar) and Tharun (Arul) are good friends and their only aim in life is to make money. They are stealing from banks, hijacking container trucks and robbing the rich people. They had been actively sought by the police, the police inspector Manikkavel (J. Livingston) is charged to catch them. The friends on the run travelled from town to town, and are later hosted by a mysterious man G. R. (K. Bhagyaraj) in his bungalow, the fugitives introduce themselves as youngsters who were lost in the wood.
Dr. Rajkumar, Kanchana, Jayamala and Padmapriya are played the main lead roles along with Vajramuni, Thoogudeepa Srinivas, Balakrishna, Sampath, Bhadrachalam, Chandrashekhar, Shivaprakash, Udayashankar, Prabhakar and many others are seen in supporting roles in this movie.
