A youngster takes revenge on four boys who killed his father.

In Mumbai, the people who had migrated from Tamil Nadu are living in the slum of Dharavi. Union Minister Haridev “Hari Dhadha” Abhayankar (Nana Patekar) tries to evict the people and seize their land. Over the years, he becomes a minister and plans to clear the area to construct buildings with the help of his supporter Vishnu Bhai (Sampath Raj). Karikaalan (Rajinikanth), living in Dharavi, opposes it to protect the area and his people. His ex-lover Zareena (Huma Qureshi) returns to Dharavi as a representative non-governmental organization that hopes to improve Dharavi’s living standards. Hari Dhadha’s assassination attempt on Kaala kills Kaala’s wife Selvi (Easwari Rao) and elder son Selvam (Dileepan). Kaala decides to take revenge on him and encourages the people to protest against the government. Hari Dhadha creates a curfew in Dharavi with the help of his supporter Inspector Pankaj Patil (Pankaj Tripathi). Kaala fights with Hari Dhadha’s henchmen and the police. Kaala’s younger son Lenin (Manikandan), daughter-in-law Charumathi “Puyal” (Anjali Patil), and brother-in-law Vaaliyappan (Samuthirakani) fight alongside him. Pankaj gets arrested after a video taken by a reporter Praveen (Ramesh Thilak) provides evidence that the henchmen were brought into Dharavi to create problems with the police’s support. The police believes Kaala is dead and informs the media, but the people of Dharavi deny the news of his death. Hari Dhadha enters Dharavi at a function for constructing a new building. Here, Kaala reappears amongst the crowd. Hari Dhadha is killed by the people of Dharavi in a stampede, and the government stops the scheme. With Dharavi’s success, many slums start to follow Kaala’s way of protest to protect their neighbourhood, irrespective of illegal land occupation.

Rohan Bhatnagar (Hrithik Roshan) is a kind, happy young man who has been blind since birth and works as a voice-over artist for a living. Through friends, he meets Supriya Sharma (Yami Gautam), a working woman who is also blind, but proudly independent. The two fall in love and get married.
One night, on their way back home after dining out, they are interrupted by Amit Shellar (Rohit Roy), a local goon and younger brother of Madhavrao Shellar (Ronit Roy), a well-known politician. He and his friend Wasim, drunkenly misbehave with the couple, enraging Rohan, and they have a heated exchange.

The story revolves around an ego clash between father and son. Puli (Sundar C) is a local rowdy and the son of a math teacher Vedhukannu (Prakash Raj). Vedhukannu hates his son for being a rowdy and not being proficient in math. As part of a flashback, it is shown that young Puli was very good in physics, but his father, being a math teacher, wanted him to excel in math, which he was unable to do. As a result of repeated physical and mental abuse, Puli takes the wrong route to grow up. In the meantime, rival gangs and police try to kill him, but he is accidentally saved by his half-sister, a daughter from an affair his father had. Puli helps his half-sister in return for her unexpected help and vouches for her safety. The rival gangs kill her and put the blame on his father. How Puli brings the truth to light and unites with his family completes the rest of the movie.

Prabha (Aadhi) is the eldest son of his family who does nothing but spend all his time with his friends. He is chided by his father (Jayaprakash) for that. Being a volleyball player comes to his rescue as he finds a part-time job as a volleyball coach in a women’s college. His younger brother works in a TV channel, and Prabha is often compared to him by the family, which incurs his wrath. The siblings cross swords with each other. One day, the younger brother is killed, and the blame falls on Prabha. He is on the run and soon joins a goonda working as a henchman to an influential politician (Mahadevan). Prabha goes on a killing spree, killing one by one in the gang. Finally, a flashback reveals that they were the reason for his brother’s death.
