The movie is set in the 1990s and follows the life of Manoharan, who is a milk vendor. Due to a tragic turn of events, Manoharan puts his whole family at risk, almost becoming an orphan in the process. This is when he comes to know about a secret, that takes him on a new journey in life.
Anjana Ali Khan announced that she was working on her second feature film in November 2014 and that the film would be a romantic comedy featuring actors Rahul Ravindran and Wasna Ahmed. The title is inspired by the 1975 Tamil film Pallandu Vazhga, with a play on words changing the title to include Palaandu (teeth) rather than Pallandu (long), to showcase the lead’s character’s occupation as a dentist. She worked on the script in Qatar, before moving to Chennai to begin the shoot. Chandini Tamilarasan stated that she was a part of the film and would be playing the role of an IT professional. Nani, who starred in Ali Khan’s debut film Veppam, shot for two days in Chennai for a cameo role.

The film is a story of two childhood friends who live their lives to the fullest. The story focuses on the friendship, love lives and careers of both the friends and how one of them sacrifices his life at the cost of his career.
The story begins with a wedding having the bride, but the groom Guru (Chaams) is missing. The bride then decides to call Guru’s friend, Billa (Vinay Rai), who does not know where Guru is. A flashback then begins.
Running away from their lives due to various problems involving them, Billa, Ranga (Sathyan Sivakumar), and Kochadaiyaan (Aravind Akash), who are good friends, decide to live a bachelor life. However, before they head off, they decide to invite their good friend Guru. Despite the fact that Guru’s wedding is in a few days time, he decides to join in the gang. The four friends then take an oath to live a bachelor’s life and not reveal the truth that they are married. When they reach the city of Bangalore, they approach Charles (Premgi Amaren), a college mate of theirs who is now a multimillionaire. Charles at first was scared of them as the four of them teased him in college. After a change of mind, he decides to help them live their bachelor’s life and bring them to a party. At the party, they see a model Sanjana (Lakshmi Rai), who danced with them. After the party, Charles brings them to a private condominium where they stay. At the condo, they see Sanjana. The four of them try their best ways to get Sanjana as their girlfriend.
Raasaiyya (Prabhu Deva) lives with his grandfather, Shyamsunder (Vijayakumar), a kind-hearted and honest man in a small town in rural Southern India. Joker is a political satire which criticises the present political absurdities. Mannar Mannan (Protagonist), a rural villager, who self declares himself as President of India, always protests Government for all the absurdities happening around his village. Most of the time he is seen as a Joker. He’s trailed at court for his various continuous acts. There comes his past, where he loves a beautiful girl from his near by village. The girl is willing to marry him only if he could build a toilet at his home. Government announces a toilet scheme through which all the villagers will get toilet at their homes throughout India. President plans to visit a village and he chose protagonist’s village. What happens when the President comes? is toilet built at their home? did they get married? answers protagonist’s act of declaring himself as President. At the end, the audiences are questioned for their involvement in opposing absurdities (who’s the real Jokers)!
